Upcharge for faster shipping ... on PRIME? Is this a new thing?

I think it's new i assume that the option with the time slot is that they use Amazon shipping service instead of USPS because with USPS it's quite hard to guarantee a time I have had in the past anywhere from 9 in the morning to 6 pm .
The only difference between a good day and a bad day is your attitude(:

Been seeing / using it for the past 2 months.
Spend over $35 on eligible items to waive that fee.

Next year they will sell us a "Prime PLUS" which will remove that $35 minimum requirement for faster shipping.

Never even noticed that option, though I have seen a few Amazon Prime branded delivery vans. If they're waiving the $2.99 for orders over $35, they should say so, it might incentivize some people to order more or combine items into one bigger order.

Considering how slow prime shipping has been (still takes quite a while for some items), that's just piggish on Amazon's part.

Likely not all cities have it, and of course not all Prime items are eligible.

Search filter available online (also available on mobile app, just harder to get to):

And how it shows by a product:

I've used it to get items "overnight, between 4AM - 8AM".

Seems like a hybrid between the regular Prime delivery vans (Amazon shipping), and Prime Now.

Considering you get regular Amazon pricing and inventory (for available items), it's a good hybrid.

At this point "Prime" shipping is meaningless, it just means free shipping. It might be next day or it might be a week and a half, either way they still call it Prime.

Generally true. But items scheduled to arrive within a day or two typically arrive on time IME, wbu?

My recollection is that Prime was never more than 2 day shipping

Yes, but
Prime was never more than 2 day shipping
It used to be that Prime meant 2 day shipping (then later they added one day.) So it used to be that Prime meant 1-2 day shipping. Nowadays they call all free shipping Prime no matter when it's scheduled to arrive. As I said, it might be one day and might be 2 weeks, but it's all called still called "Prime."

On the bright side, at least they're only calling it "Prime" now, for a while they were still calling it "2 day" even if it took a week because "it's 2 days from when it ships." I have screenshots where within one order one item would have two day shipping one day and another item would have what was labeled the same "FREE Two-Day Shipping" several days later. At least they stopped that scam.

Just today I almost spent more than $100 extra to buy something from a store that would guarantee 10:30am shipping as opposed to Amazon that didn't offer that option (in the end it ran out of stock before I completed the order).

More options is always great.

$100 extra on what price? If it's $100 extra on a $5000 item that you really need right away, then it could easily be worth it.

Nah. There are many alternatives ranging the gamut from none to depends. Nothing justifies paying $100 for shipping underwear.
I've been waiting over 5 years with bated breath for someone to say that!
