Try to find something with SPG Starpoints, Cash & Points shouldn't be too expensive and @ SPG he might get a decent upgrade for his wedding night (email the manager in advance with a PDF of the invitation).

Does anyone have experience with this? I used click-to-call to make the reservation, and the rep told me that she put a note in to make an upgrade because they're newlyweds, wedding night and yadayadayada. Is there any reason to still reach out to the manager?

Of course there is -- as the wedding gets closer see if they can confirm you into an upgraded room, so you'll know for sure.
