I'm looking to book a one way, non-stop from TLV to JFK for 45k. Can I expect one to open up in the future if it hasn't in the past month or two?

TLV-EWR is the correct rout for United

If I have MR, how would I best book a UA domestic flight - transfer to SQ or ANA?

Negative. That thread is probably the least responded thread on DDF. I know how to book normally, now I'm looking for the tricks like transferring to a partner that requires less miles than UA itself.

Well then it depends from where to where do you plan on traveling?

What I understand is that ANA uses fewer points, is only round trip ( workaround available), and takes longer for the MR transfer to happen.  I don't recall their cancellation policies.
Both won't have have fuel surcharges.

Domestic US economy will be 12.5k Aeroplan miles with no close in fee for a OW.

The beauty of SQ and LH is when redeeming for J.

Domestic - coach. Within that, I would think it shouldn't matter.

Thanks! Again, though, since it's domestic, there won't be YQ anyway just the $5.60 fees. Although, ilherman just pointed out, it'll be the same 12.5K as UA...

So same as UA. (No issue of close in fees here.)
Gotcha about the J. Thank you!

With UA any flights within 500 miles IIRC is just 10k miles.

Thanks. The flight is EWR-FLL (happy, Dav? ;) ) so it's 12.5K

Now to use MR, I would have to transfer to ANA? That's better (faster?) than to SQ?

SQ is faster. Also, I don't think ANA will be cheap for domestic flights.

My question applies to both NY/NJ.

Are you specifically asking about united?
