I think all the things we did were the must sees, my order would be:
1) walking tour of Old Havana and soaking in the culture, people and city streets;
2) City tour in classic convertible;
3) Cigar factory;
4) Havana music and night life;
5) Cuban Missile Crises Monument; and
6) Some of the historical and political monuments, plazas and sights.
