It depends on whether you read it as one word or as an abbreviated form of two words.  If it's one word then the second vowel (in this case the E) modifies the A, like in the words lame, tame, name etc.. If it's Am-Ex then the a is pronounced Am- like the beginning of American.

I pronounce it the #2 way too. I just wanted to offers the logic behind both options.

I called Amex and spoke to the supervisor, and the supervisor's supervisor, etc.,  up up up, all the way to the guy who doesn't realize he is giving away free vacations to the whole world, and asked him.
He said he never shortens American Express to any vulgar nicknames.
