We stayed here - https://www.booking.com/hotel/it/cagliarifornia.html - centrally located 5 minutes walk to train station and in the center of the city.

If I will be going from Venice to Tuscany (not Florence, but rather wine tours, small towns, outdoor activities, etc.) and then to Rome, am I better off:

1. renting a car from Venice and returning in Rome
2. taking a train from Venice to Florence, renting from there, return to Florence, and take train to Rome
3. taking a train from Venice to Florence, renting from there and returning in Rome

Any recommendations?

Driving all the way from Venice isn't necessary.  The trains are fast and efficient.  (If you book earlier, it's cheaper.)
#3 saves you taking a train from Florence to Rome. Saves time, but:
Compare to the drop off fees for a 1 way rental.
