As i once did this and paypal ended up calling the shot.

Did you get the first item? If yes try to dispute that too, saying it was a package deal and he defrauded you. Teach him a lesson and come out ahead.

If Amex needs proof, you have correspondence that he agreed to sell you an item and that you will pay via PP.  Your claim is that you never received the item.

I don't know if you will have any luck if PayPal wants to fight Amex.

The PayPal transaction was valid, since it was sent as a gift. Even though you have email correspondences.

I opened a eBay store last week to get discounts on the fees, however I sold a a few items and I didn't see any discounts, so she tells me that because I LISTED the item before opening the store, I will not get the discount. Is this true, if yes will end listing and relist help?

I also don't know the exact answer. It does make sense that it goes from when you listed the items.
