Off the cuff thoughts.
Covid will continue to be with us. So the vaccine will continue to be relevant.
Although the USA has a high vaccination rate, there are many people around the world who are waiting for a jab.
There are probably other considerations, but I would expect  it to continue to rise or at least hold it's own for a while longer. 
At some point it will level off and it may be a long term hold, or a sell.
I don't see it dropping drastically,  unless something really big happens.  So you may want to hedge by selling just enough to cover the original cost, so you'd have no capital loss in case of sudden disaster.
 
And congrats on getting in so early!

wow it went from around a $7b market cap before the pandemic to now having a $125b market cap

Keep in mind it is a very new company (founded in 2010) and they currently only have 1 product although they are working on others.

my opinion is it will take at least 2 years for the world to have herd immunity and I wouldn't be surprised if they recommended yearly booster shots too, so they should have plenty of business for the foreseeable future.

My problem is that for a pharmaceutical company that is ~10 years old and currently only has 1 EUA product, to be valued at half the value of a company like Pfizer ($224b mkt cap) which has been around for 170 years (which also has a similar vaccine) just doesn't make sense.

$MRNA might continue rising but I wouldn't put my money into it, at this point the valuation doesn't make sense to me.

All these points are great but no one can predict the future, let price lead you @yas. I would suggest a trailing stop loss order on a percentage you feel comfortable and forget about it.

At the current price, Selling a third of your shares leaves you only profits to be in.  Maybe do that.  So you stay for some upside and your original principal is out.  It's a comfortable way of investing

Theres absolutely no reason to sell, with any stop loss order you can protect yourself from downturn. Why sell ?

The only reason I can think of for not doing as you suggest is if you think the returns from here on out are minimal (which may be the case as per @Shmulie's post), and there are better uses or opportunities elsewhere.

So why won't a stop loss retain those profits? By selling enough to regain his capital and continuing to risk the profit by letting it ride, he's not retaining profit; he's just ensuring he doesn't go into the red.

Yes if you are a trader looking to utilize cash elsewhere. but from what it seems for @yas its a one time trade and looking for advise on how to best keep it secure, in which case as you agreed with me  ;) a stop loss or trailing on the amounts you think are a pullback is the best course.

A stop loss order doesn't protect you from disaster.
If there's big news and the stock drops quickly, maybe even opens at significantly down from yesterday's close, your stop orders cashes you out at what ever the price is.

The probability of that happening is pretty low IMO, whereas the continued upside is higher
